2.2.6. Micropestle

You will need individual disposable micro pestles that fit microcentrifuge tubes. These can be bought or made by heating a 1000 μl blue tip in a flame and moulding it into a disposable pestle in a microcentrifuge tube while it cools.

  • Pros: Single bees; cheap; low maintenance.
  • Cons: Time consuming; lack of uniformity.

  1. Grind a frozen bee tissue or larval sample with the micropestle in a microcentrifuge tube.
  2. Discard pestle.
  3. Add 500 μl buffer per bee.
  4. Mix with a vortex.
